Got potentially 300-400 sign panels to supply.

Needs to be 40" x 72"

Indoor use needs to be 0.21 thick at least.

What are the options to get prices competitive?

We have UV printer.

Any thoughts would really appreciated

Thanks!

Styrene. But the size isn't great for material utilization, not sure of you can get styrene in rolls like polycarbonate?

6mm PVC/Sintra.
.25in MDF or Hardboard
6mm Coroplast

Without knowing anything about the project, I'd imagine 6mm PVC would be the way to go.

Roper of Goats. Old ones.
styrene would be limitied lifetime -- becomes brittle and starts to crack and yellow -- yes, even indoors

Sintra (or similar) if surface needs to be clean-able

FomeCor or similar if panels will be away from the possibility of being touched.

Contact a substrate manufacturer directly (depending on which material you want to use) and ask them for a custom quote. For example, corrugated plastic is an extruded product. The manufacturer can cut any length they want, and trim the width to size. Should be very cost-effective (not much waste), and you won't have to cut down anything in-house.
