I have a 12"x96" 2nd surface printed acrylic that I am mounting to a 12"x96" painted MDF backer. What is your opinion on the best adhesion method?

Clean the backside with isopropyl alcohol,.
With a good dual side mounting adhesive such as the Neschen range (GUDY 802, 831,...) or similar aqueous/ acrylic based glue, it should work fine.
