Need Help Colors ok on test print, but don't show up when printing.

I have a Roland VP 540 and when I do a test print the cyan and yellow show up, but have some issues, but when I run a print the yellow shows, but the cyan doesn't. I am new to this printer so I don't know much, but thus far I have manually cleaned the print heads, ran a few small, medium and heavy cleans and even soaked the print head for a few hours. Does anyone have any suggestions on what is causing this or what else I may do?
