Create your setup as right readng with all colors except the white set to overprint. In GSP Plot set the job to reverse reading and also reverse the print order. It should print fine following that approach.

You're wasting your time searching for a file format that will work. FlexiSign, in the version you're running, does not support transparent backgrounds. So PNG, GIF and TIF with transparency will still be seen by Flexi as having a background. It is entirely possible that the extra auto tracing...
