You can use production manager to "Tile" the graphic and select .5" as your overlap.

If you're using Illustrator you'll want to use the CadTools plugin from Hot Door. It has a fillet tool that will round your corners. Otherwise you could always use Flexi's "Round Corner" tool and select the "Inside Corner" option.

I don't have my Flexi key here at home so I'm unable to open PM to see if there is a setting to allow you to print the substrate color. It may be easier to just draw a shape the same size of the substrate, set the color of that, then send it to the back of your design.
