To me it looks like something stuck to the bottom of your carriage. Could be static issue but doesn't look like it. Either that or your material is buckling in the print area and the print carriage is touching it as it passes over.
Test prints. Pictures of a print? Sounds like contamination either from inside the printer as it goes through or wherever the media is sitting before printing.
I would try factory resetting your plotter.
Turn off. Turn on and quickly hold up and enter. Go to set defaults. Look at the language. Reset. Set language back to what it was. Then power off and on again and try your cut again.

If it's vector still then it should print fine. YOu can always send the whole file and then just print one 54" x6" section to print. That way the area with issues is getting printed without printing the whole thing. I would try that see. If you have a vector in illustrator and save as .jpg...
