The only way to print on clear with opaque qualities is to back print with white ink. Or print on clear, apply to white vinyl and contour cut. As far as I know anyway.

Probably did not suck air into the line. If the cartridge is empty, or very near, it can hold a vacuum on the ink in the lines and prevent flow of think to the head. Replace the cartridge, perform normal clean and proceed. Keep us posted.

For testing.... clean a section of material with denatured alcohol, let dry and then print on it. See if you get the same dots. Looks environmental to me.
